چکیده
naling pathway. For this, we focused on Pea3/Ets (Pea3, Erm, Er81), a subfamily of Ets transcription factors, since it has been suggested that Pea3/Ets is a direct transcriptional target of Fgf signaling. We first checked that Pea3 and Erm are expressed in the midbrain and the hindbrain around stage10 of the chick embryos. We then obtained results that Pea3 and Erm expressions are regulated by Fgf-Ras/ERK signaling; Pea3/Erm were induced after misexpression of Fgf8 and repressed by misexpression of dominant-negative form of Ras. In order to examine function of Pea3/Erm in the isthmus, we constructed dominant-negative form of Pea3 (Pea3-DN) and misexpressed it in the mesencephalon and the metencephalon at st.9 chick embryos. Surprisingly, Shh was ectopically induced at Pea3DN expressing sites in the midbrain and hindbrain. Oculomotor neurons that differentiate in the ventral midbrain differentiated ectopically in the dorsal side of the midbrain. It is speculated that Fgf signaling is involved in not only caudorostral axis formation, but also ventro-dorsal axis formation of the midbrain by repressing Shh through Pea3/Ets transcription factors.

Shh, Bmp-2, Bmp-4 and Fgf-8 are associated with initiation and patterning of mouse tongue papillae
Spacing patterns are of fundamental importance in various repeated structures which develop at regular intervals such as feathers, teeth and insect ommatidia. The mouse tongue develops a regular papilla pattern and provides a good model to study pattern formation. We examined the expression patterns of the signalling molecules, sonic hedgehog (Shh), bone morphogenetic proteins -2 and -4 (Bmp-2 ...
متن کاملFgf9 signalling stimulates Spred and Sprouty expression in embryonic mouse pancreas mesenchyme.
Epithelial-mesenchymal interactions are critical for normal pancreas development. Fibroblast growth factor (Fgf)-10 is expressed in the pancreatic mesenchyme and its signalling is required for normal growth and regulation of gene expression in the pancreatic epithelium. However, little is known about putative Fgf signalling to the mesenchyme. Here we have examined the embryonic pancreas express...
متن کاملFGF signalling is required for differentiation-induced cytoskeletal reorganisation and formation of actin-based processes by podocytes.
To examine the potential role of fibroblast growth factor (FGF) signalling during cell differentiation, we used conditionally immortalised podocyte cells isolated from kidneys of Fgf2 mutant and wild-type mice. Wild-type mouse podocyte cells upregulate FGF2 expression when differentiating in culture, as do maturing podocytes in vivo. Differentiating wild-type mouse podocyte cells undergo an epi...
متن کاملRetinoic acid orchestrates fibroblast growth factor signalling to drive embryonic stem cell differentiation.
Embryonic stem (ES) cells fluctuate between self-renewal and the threshold of differentiation. Signalling via the fibroblast growth factor (Fgf)/Erk pathway is required to progress from this dynamic state and promote mouse ES cell differentiation. Retinoic acid also induces differentiation in many cellular contexts, but its mechanism of action in relation to Fgf/Erk signalling in ES cells is po...
متن کاملRoles of Wnt8a during formation and patterning of the mouse inner ear
Fgf and Wnt signalling have been shown to be required for formation of the otic placode in vertebrates. Whereas several Fgfs including Fgf3, Fgf8 and Fgf10 have been shown to participate during early placode induction, Wnt signalling is required for specification and maintenance of the otic placode, and dorsal patterning of the otic vesicle. However, the requirement for specific members of the ...
